Use of Randomized Submaximal Glutamate Stimulus to Interpret Glial Effects on Neuronal Calcium Dynamics



The neuronal synapse is a dynamic functional unit which is composed of neurons and support glial cells, which provide the important function of glutamate re-uptake. We have previously described calcium dynamicsin neuronal cultures treated with toxic and non-toxic concentrations ofglutamate using single stimuli of micromolar glutamate . Here we investigated whether a pattern of multiple, submaximal glutamate stimulations at nanomolar concentrations of glutamate affected calcium dynamics in neuronal cultures with and without high glial content, and whether the order of addition of nanomolar glutamate affected calcium dynamics. Calcium is a key signaling ion involved in memory and learning with ionotropic glutamate receptors such as the N-methyl-D-aspartate receptors (NMDAR) on the neuronal membrane. 

Glial Effects on Neuronal Calcium Dynamics
 NMDARs are a major subtype of ionotropic receptors responsible for binding glutamate, the most abundant excitatory neurotransmitter (excitatory stimulus) in the human brain. Activation of NMDARs opens the ion channel at theplasma membrane to allow calcium influx into the cell cytosol. Calcium in its ionic form is very dynamic, especially in excitable cells such as muscle and brain cells, moving from the high concentration exterior of the cell to the much lower concentrations inside the cell where calcium is used as a second messenger.

The permanentpresence of these unrecognized resources in the urban environment constitutes athreat, causing several diseases. Thus, resulting in the decrease in life expectancy of human population. Interest in waste management is evident and is observing in several researches, but also by the proliferation of international meetings of reflection revolving mainly around the environment and safety.

Stockholm 1972, Kyoto 1977, Nairobi 1991, Rio de Janeiro 1992 and Johannesburg 2002 summits can be considered as those used engine about the awakening global consciousness on environmental issues.Through various efforts around the world, it appears thatthe sanitation level is especially improving in developed countries. The case of some European countries that minimize spread risk of diseases that typically generate the excreta through the drain, transportation and disposal of sludge can be evoked.

Combination of ESI and MALDI-MS Imaging Enhances Analysis on Brain Gangliosides



Recent advances in matrix-assisted laser desorption ionization (MALDI) mass spectrometry imaging (MSI) have led to the direct analysis of tissue slices. The major advantage of MSI is its capability of simultaneously localizing and identifying a parent molecule and its metabolites without any labeling or any prior knowledge. MSI has been extensively employed to detect the differentiated pattern of lipids in various organs in different diseases, such as brains in Alzheimer’s disease. Poor reproducibility of MALDIMSI analysis due to the heterogeneity of the matrix-analyte crystals, hindersits use on quantitative analysis. In addition, discontinuous ion flow due to quickly consumption of the samples under laser irradiation on specific site affects its ability in qualitative analysis. Although electrospray ionization mass spectrometry (ESI-MS) cannot directly be used for imaging, ESI tandem mass spectrometry (LC/ESI-MS/MS) can separate and distinguish gangliosides.

Combination of ESI and MALDI-MS Imaging

Zhang et al. have presented a promising workflow for qualitative, semi-quantitative and in situ analysis of gangliosides bycombining the MALDI MSI and ESI-MS. Following obtaining the brain from the mice, fresh-frozen murine brain sections were prepared and coated with matrix for subsequent MALDI MSI analysis.

Chromium(VI) Removal Using Biosorbents Derived from Moringa Oleifera



Many coagulants are widely used in conventional water treatment processes based on their chemical characteristics. These coagulants are classified into inorganic, synthetic organic polymers and natural coagulants. Coagulation is the most essential process in the treatment of both turbid surface and industrial wastewaters. Coagulationflocculation is one ofthe most important physicochemical treatment steps in wastewater treatment toreduce the turbidity of wastewater. Due to the lack of proper water treatment systems in these rural or under developed communities the best immediate option is to use simple and relatively cost effective point of use technologies such as coagulation.

Chromium(VI) is highly toxic and soluble in water and it is a strongoxidizing agent that causes severe damage to cell membranes. The mostcommonly used primary chemical coagulants, namely alum (AlCl3), ferric chloride(FeCl3) and polyaluminium chloride (PAC). The disadvantages associated with these coagulants such as ineffectiveness in low temperature water relatively high procurement cost, detrimental effects on human health, production of large sludge volumes and the fact that they significantly affect pH of treated water. There is strong evidence linking aluminium based coagulants to the development of Alzhemier’s disease in human beings.

Non-Classical Progenitor Mononuclears in Metabolic Syndrome: The Role of Serum 25-Hydroxyvitamin D3



Metabolic syndrome (MetS) is a cluster of cardiovascular (CV) risk factors, including central obesity, glucose homeostasis, insulin resistance, hypertension and atherogenic dyslipidemia, appears to have anincreased prevalence worldwide. Recent clinical and observation trials have shown that the patients with MetS exhibit near 2-fold increased risk of CV disease and events, asymptomatic atherosclerosis, all cause and CV death rate.

 Serum 25-Hydroxyvitamin D3


There are evidence regarding the pivotal role of vitamin D in pathogenesis of CV disease, diabetes mellitus, obesity, and MetS. Thereliable indicator of body vitamin D status is serum concentration of25-hydroxyvitamin D [25(OH)D3]. Low serum 25(OH)D3 levels have been directly linked to MetS, whereas the overall risk of MetS in general population was not probably associated with 25(OH)D3 concentration.
